Dendritic cell stimulation by mycobacterial Hsp70 is mediated through CCR5

An effective host immune response to mycobacterial infection must control pathogen dissemination without inducing immunopathology. Constitutive overexpression of mycobacterial heat shock protein (myHsp70) is associated with impaired bacterial persistence, but the immune-mediated mechanisms are unknown. We found that myHsp70, in addition to enhancing antigen delivery to human dendritic cells, signaled through the CCR5 chemokine receptor, promoting dendritic cell aggregation, immune synapse formation between dendritic cells and T cells, and the generation of effector immune responses. Thus, CCR5 acts as a pattern-recognition receptor for myHsp70, which may have implications for both the pathophysiology of tuberculosis and the use of myHsps in tumor-directed immunotherapy.     

Triple-bond reactivity of diphosphorus molecules

We report a mild method for generating the diphosphorus molecule or its synthetic equivalent in homogeneous solution; the P2 allotrope of the element phosphorus is normally obtained only under extreme conditions (for example, from P4 at 1100 kelvin). Diphosphorus is extruded from a niobium complex designed for this purpose and can be trapped efficiently by two equivalents of an organic diene to produce an organodiphosphorus compound. Diphosphorus stabilized by coordination to tungsten pentacarbonyl can be generated similarly at 25 degrees C, and in this stabilized form it still efficiently consumes two organic diene molecules for every diphosphorus unit.     

Silk genes support the single origin of orb webs

The orb web is a spectacular evolutionary innovation that enables spiders to catch flying prey. This elegant, geometric structure is woven with silk fibers that are renowned for their superior mechanical properties. We used silk gland expression libraries to address a long-standing controversy concerning the evolution of the orb-web architecture. Contrary to the view that the orb-web design evolved multiple times, we found that the distribution and phylogeny of silk proteins support a single, ancient origin of the orb web at least 136 million years ago. Furthermore, we substantially expanded the repository of silk sequences that can be used for the synthesis of high-performance biomaterials.     

Seroprevalence of feline leukemia virus and feline immunodeficiency virus infection among cats in North America and risk factors for seropositivity

OBJECTIVE: To determine seroprevalence of FeLV and FIV infection among cats in North America and risk factors for seropositivity. DESIGN: Prospective cross-sectional survey. ANIMALS: 18,038 cats tested at 345 veterinary clinics (n=9,970) and 145 animal shelters (8,068) between August and November 2004. PROCEDURE: Cats were tested with a point-of-care ELISA for FeLV antigen and FIV antibody. A multivariable random effects logistic regression model was used to identify risk factors significantly associated with seropositivity while accounting for clinic-to-clinic (or shelter) variability. RESULTS: 409 (2.3%) cats were seropositive for FeLV antigen, and 446 (2.5%) cats were seropositive for FIV antibody; 58 (0.3%) cats were seropositive for infection with both viruses. Multivariable analysis indicated that age, sex, health status, and cat lifestyle and source were significantly associated with risk of seropositivity, with adults more likely to be seropositive than juveniles (adjusted odds ratios [ORs], 2.5 and 2.05 for FeLV and FIV seropositivity, respectively), sexually intact adult males more likely to be seropositive than sexually intact adult females (adjusted ORs, 2.4 and 4.66), and outdoor cats that were sick at the time of testing more likely to be seropositive than healthy indoor cats (adjusted ORs, 8.89 and 11.3). CONCLUSIONS AND CLINICAL RELEVANCE: Results suggest that certain characteristics, such as age, sex, health status, and lifestyle, are associated with risk of FeLV and FIV seropositivity among cats in North America. However, cats in all categories were found to be at risk for infection, and current guidelines to test all cats at the time of acquisition and again during illness should be followed.     

Ventricular pneumocephalus and septic meningoencephalitis secondary to dorsal rhinotomy and nasal polypectomy in a dog

CASE DESCRIPTION: A 4-year-old sexually intact female French Bulldog was evaluated because of lethargy, anorexia, and chronic rhinitis-sinusitis. The dog had nasal discharge of 18 months' duration; dorsal rhinotomies were performed 3 months and 2 weeks prior to referral. CLINICAL FINDINGS: On initial evaluation, intraventricular pneumocephalus and sinusitis were diagnosed; CSF analysis revealed high total protein concentration and mononuclear pleocytosis. The dog's condition improved with treatment. Two weeks after discharge, it was treated by a local veterinarian because of upper airway obstruction; 3 days later, the dog was referred because of seizures. Computed tomography revealed a large fluid-filled, left lateral ventricle and a soft tissue mass protruding through a cribriform plate defect. The mass was histologically consistent with brain tissue. Findings of clinicopathologic analyses were unremarkable. Results of cytologic examination of a CSF sample were indicative of septic, suppurative inflammation, and bacteriologic culture of CSF yielded Escherichia coli. TREATMENT AND OUTCOME: Amputation of the herniated olfactory bulb and antimicrobial treatment resolved the septic meningoencephalitis, but neurologic deficits recurred 6 weeks later. Definitive correction of the cribriform plate defect with bone and fascial grafts was attempted. Postoperative rotation of the bone graft resulted in cerebral laceration and hemorrhage, and the dog was euthanized. CLINICAL RELEVANCE: Findings suggest that following dorsal rhinotomy and nasal polypectomy surgery, the dog developed herniation of the left olfactory bulb, intra-ventricular pneumocephalus, and septic meningo-encephalitis because of a cribriform plate defect. Care must be taken to prevent rotation of bone grafts used in cribriform defect repair.     

A retrospective investigation of feline gastrointestinal parasites in western Canada

Between 1998 and 2008, feline fecal specimens were submitted to provincial veterinary diagnostic laboratories in Regina and Saskatoon, Saskatchewan, for sucrose centrifugation-flotation (n = 635), parasite identification (n = 17), and/or Giardia (n = 283) or Cryptosporidium (n = 266) commercial direct immunofluorescence assay (IFA). The most commonly detected parasites on flotation were Toxocara cati (4.7%), Isospora (3.8%), and taeniid eggs (Echinococcus or Taenia) (1.3%). Cats less than 2 years of age were twice as likely to have a positive parasite test as cats older than 2 years. Using IFA, Giardia was detected in 9.9% of samples, and Cryptosporidium in 2.3% of samples. Relative to IFA, flotation had sensitivity values of 39% and 50% for detection of Giardia and Cryptosporidium, respectively. Giardia and Isospora were detected in a higher proportion of samples in our study population than reported in the general cat population in western Canada. This study highlights the importance of sensitivity when interpreting diagnostic tests and provides information to guide region-specific recommendations for helminth parasite prevention and treatment.
